Human skin is a complex bilayered organ that serves as a protective barrier against the environment, and the loss of its integrity due to traumatic experiences can lead to severe consequences. This review aims to investigate the potential of silk, particularly silk fibroin, as a biomaterial for dermal constructs necessary for skin regeneration. A comprehensive analysis of randomized-controlled trials was conducted, focusing on current dermal constructs and the unique structures and properties of silk-based constructs on wound healing. The findings indicate that silk fibroin is regarded as the most promising biomaterial, providing significant options for constructing tissue-engineered skin. This research underscores silk fibroin's suitability as a scaffold to create adequate dermal constructs, which are essential for optimal recovery from skin injuries. The review highlights the mechanical and biological properties of silk proteins, sericin and fibroin, emphasizing their role in enhancing the efficacy of dermal substitutes. Moreover, it elaborates on the necessity of developing effective cutaneous substitutes to restore the functional properties of native skin.
