This investigation evaluated doxorubicin-induced hepatotoxicity and the preventive effect of green-synthesized magnesium nanoparticles (ObE-MgNPs) derived from Ocimum basilicum in rats. Bioactive molecules were extracted and qualitatively screened phytochemically, and green synthesis of MgNPs was confirmed by visual color change, FT-IR spectroscopy, and SEM analysis. Fifteen female albino Wistar rats were divided into three groups: control, doxorubicin-treated (DOX), and doxorubicin plus MgNPs co-treated (DOX + MgNPs), and various hematological, enzymatic, and oxidative stress markers were measured. FT-IR analysis confirmed the formation of MgO nanoparticles, and SEM revealed mainly spherical particles with an average size of about 78.5 nm; phytochemical screening showed Ocimum basilicum extract to be rich in phenolic and other secondary metabolites. Doxorubicin administration altered hematological parameters, increased liver enzyme activities, and induced oxidative stress in liver tissue compared with controls. Co-treatment with MgNPs partially reversed these alterations and improved oxidative status. The results suggest that the antioxidant properties of magnesium nanoparticles confer hepatoprotective effects against doxorubicin-induced toxicity and may be useful in mitigating chemotherapy-related organ damage.
