This study investigates the acid phosphatase activity during the differentiation of the chick liver, utilizing both histochemical and quantitative biochemical methodologies. It was found that the activity levels of acid phosphatase are not static; rather, they fluctuate significantly at various stages of liver development. Elevated levels of activity were observed during the first eight days of incubation, followed by a decrease until fourteen days, after which activity levels increased again in later developmental phases. The investigation revealed that higher acid phosphatase activity is present in the perivascular connective tissue compared to the parenchymal cells during the incubation period of eight to twenty days. Additionally, a notable distinction in enzyme activity was found in early developing red blood corpuscles, where nuclear activity was prominent, shifting to cytoplasmic predominance in advanced stages. This fluctuation in acid phosphatase activity is suggested to correlate with cellular death and catabolic processes involved in chick liver morphogenesis. The findings offer insights into the relationship between enzyme activity and the growth and differentiation of the liver.
